The family of ryanodine receptor (RyR) genes encodes three Ca2+ release channels: RyR1, RyR2 and RyR3. In addition to their well known role in regulating contraction in striated muscles, RyRs are expressed in many other cell types, where eventually multiple RyR isoforms can be co-expressed. Recent studies have revealed that several important regulatory mechanisms can modulate RyRs activity under normal and pathological conditions. In this review we shall summarise the most recent developments in this area of research.
