The freeze-etching method has been applied for the ultrastructural investigation of human blood platelets. The results are in agreement with the data from animal platelets. The platelet plasma membrane shows a large number of randomly dispersed small particles with diameters of 5-15 nm. Moreover, there exist invaginations on the plasma membrane with a diameter of 30-150 nm called protuberances which seem to represent the morphological aspect of exocytotoxic processes. In the cytoplasm α-granula, dense bodies, open canalicular systems, which are in contact with the plasma membrane, dense tubular system, glycogen, Golgi-apparatus and sometimes microtubules, microfilaments and mitochondria can be distinguished. The practicability of this method for the investigation of pathological platelet membrane alterations is discussed.
